The mechanism
How it works.
Dulaglutide is a once-weekly GLP-1 receptor agonist made by fusing a GLP-1 analog to a fragment of an antibody, which makes the molecule large enough to clear slowly and extends its half-life to days. It activates GLP-1 receptors, which prompts glucose-dependent insulin release, lowers glucagon, slows gastric emptying, and acts on appetite pathways in the brain. Those effects have been studied mainly in type 2 diabetes, where they lower blood sugar and produce modest weight loss.
Safety & regulatory boundary
The consequential part.
Dulaglutide carries a boxed warning for thyroid C-cell tumors based on rodent data and is contraindicated with a personal or family history of medullary thyroid carcinoma or MEN 2. Labeling covers pancreatitis and gastrointestinal effects. Trulicity is approved for type 2 diabetes and cardiovascular risk reduction, not for weight loss, and it is not the same as compounded or research-labeled dulaglutide.
