FOXO4-DRI is an experimental senolytic peptide designed to kill senescent cells, the worn-out cells that accumulate with age and drive inflammation. It works by blocking the interaction between the FOXO4 and p53 proteins that keeps these cells alive. In aged mice it cleared senescent cells and improved some measures of fitness, kidney function, and fur regrowth. There are no human trials. The Watchlist boundary is keeping this to its mouse data and not letting an early anti-aging laboratory result become a product claim.

The mechanism

How it works.

FOXO4-DRI is an experimental senolytic peptide built as a D-retro-inverso sequence, meaning it is made from mirror-image amino acids in reverse order so cellular enzymes cannot easily break it down. Senescent cells stay alive partly because the FOXO4 protein binds p53 and holds it in the nucleus, keeping it from triggering cell death. FOXO4-DRI mimics part of FOXO4 and competes for that binding, releasing p53 so the senescent cell kills itself, while largely sparing healthy cells. This selectivity is what produced the improvements in aged mice, but the mechanism rests on rodent and cell-culture work and has not been shown to operate this way in people.

FOXO4-DRI is not an approved drug and has not been tested in published human trials. Any product sold under this name is an unapproved research chemical of uncertain identity and purity. Senolytic strategies are biologically powerful and could harm normal cells or healing if misapplied, and none of this has been characterized in people for this peptide.
