MK-677 (ibutamoren)

Also: ibutamoren · MK-0677 · GH secretagogue

MK-677 (ibutamoren) is an orally active growth hormone secretagogue that raises growth hormone and IGF-1 by acting on the ghrelin receptor. It has been studied in several human trials but is not an approved drug, and it is widely sold in the research and supplement gray market. The Watchlist boundary is separating its real but mixed human evidence from broad anti-aging and muscle claims, and noting its effects on blood sugar and fluid.

The verdict

Our read at a glance.

Whether MK-677's ability to raise growth hormone and IGF-1 translates into the muscle, anti-aging, and performance benefits commonly claimed for it.

The signal
Randomized trials show MK-677 raises growth hormone and IGF-1 and can increase lean mass and fluid, with mixed results on function and clear effects on appetite, blood sugar, and water retention. It was studied for conditions such as frailty and hip fracture but was not approved for them.
The unknown
Whether raising growth hormone with MK-677 produces meaningful long-term benefits rather than mostly water and biomarker changes, and how its glucose and other risks balance against any benefit.
Our read
Real GH-raising activity, thin proof of benefit. MK-677 reliably increases growth hormone and IGF-1, but the human evidence for meaningful muscle or anti-aging benefit is limited, and its glucose and fluid effects plus its unapproved gray-market status are the real story.

The mechanism

How it works.

MK-677 (ibutamoren) is a non-peptide molecule that mimics the hunger hormone ghrelin. Taken by mouth, it binds the ghrelin receptor (GHSR) in the pituitary and hypothalamus and triggers the gland to release growth hormone in natural-looking pulses, which in turn raises IGF-1 in the blood. Because it copies the body's own GH-release signal, it reliably moves those hormones, but this established human pharmacology is about the hormone change itself and does not by itself prove the muscle or anti-aging benefits often claimed.

Safety & regulatory boundary

The consequential part.

MK-677 is not an approved drug. It can raise blood sugar and reduce insulin sensitivity, cause fluid retention, and it failed to gain approval in the indications it was studied for. It is widely sold as a research chemical or in the supplement gray market, where product identity and quality are uncertain. Raising growth hormone is not free of risk, and its biomarker effects do not validate anti-aging or performance claims.

Context

Why this is discussed.

MK-677 is heavily marketed for muscle, sleep, and anti-aging because it reliably raises growth hormone, and it is one of the most-discussed gray-market performance compounds.
